RENAL REGULATION OF NACL, WATER AND ACID-BASE BALANCE

Abstract :
The long term goal of our research is to define the mechanisms by which the proximal tubule of the mammalian kidney transports sodium, bicarbonate, chloride and water and to characterize the manner by which they are physiologically regulated. To achieve this goal multiple techniques (free-flow micropuncture, in situ microperfusion, in vitro microperfusion, isolated membrane vesicles) are used and the data obtained by these various means are evaluated for internal consistency and integrated into a physiologically meaningful framework. Specific aims are: 1) To further characterize the nature of the sodium-dependent and sodium-independent components of acidification. 2) To examine the role of adaptive changes in Na/H antiporter activity in modulating the rate of acidification. 3) To examine the role of sodium-dependent and sodium-independent mechanisms in the regulation of cell pH. 4) To determine the response of cell pH to changes in luminal and peritubular bicarbonate, Pco2 and pH. 5) To examine the role of cell pH in the regulation of the rate of acidification in response to changes in acid-base and potassium balance, and the administration of various agents such as glucocorticoids or parathormone. 6) To further characterize the luminal and basolateral mechanisms for transcellular Na and Cl transport. 7) To obtain more precise estimates of the transepithelial driving forces for water movement in vivo.

Abstract :
We plan to continue our studies on acidification on reabsorption of NaCl and water in the proximal convoluted tubule of rat and rabbit kidneys. With respect to the mechanism of acidification we plan to: 1) examine the nature of the ion (H ion, HCO minus over 3, OH minus) across the luminal and basolateral membranes, 2) identify and quantify the pathways for bicarbonate backleak from blood to lumen, 3) define the specific roles of carbonic anhydrase present in the cytoplasm and in luminal and basolateral membranes, 4) develop a kinetic model for acidification, and 5) examine the mechanism by which homeostatic regulatory factors exert their effects. In the second part of the proposal we plan to examine the mechanism by which NaCl and water are reabsorbed, the role of organic solutes (e.g., glucose, amino acids, HCO minus over 3), and anion concentration gradients in this reabsorptive process, and the manner in which certain regulatory factors (e.g., peritubular oncotic pressure, acid-base status, potassium, PTH, etc.) influence the rate of this reabsorptive process.

Abstract :
We plan to continue our studies on acidification on reabsorption of NaCl and water in the proximal convoluted tubule of rat and rabbit kidneys. With respect to the mechanism of acidification we plan to: 1) examine the nature of the ion (H ion, HCO minus over 3, OH minus) across the luminal and basolateral membranes, 2) identify and quantify the pathways for bicarbonate backleak from blood to lumen, 3) define the specific roles of carbonic anhydrase present in the cytoplasm and in luminal and basolateral membranes, 4) develop a kinetic model for acidification, and 5) examine the mechanism by which homeostatic regulatory factors exert their effects. In the second part of the proposal we plan to examine the mechanism by which NaCl and water are reabsorbed, the role of organic solutes (e.g., glucose, amino acids, HCO minus over 3), and anion concentration gradients in this reabsorptive process, and the manner in which certain regulatory factors (e.g., peritubular oncotic pressure, acid-base status, potassium, PTH, etc.) influence the rate of this reabsorptive process.

CONFERENCE ON HYDROGEN ION TRANSPORT IN EPITHELIA

Abstract :
An International Conference on Hydrogen Ion Transport in Epithelia has been organized as a special satellite symposium of the XXIX congress of the International Union of Physiological Sciences and will be held in conjunction with the Fall Meeting of the American Physiological Society in Honolulu, Hawaii, August 21-24, 1983. The purpose of the Conference is to provide a forum for scientists working on various aspects of hydrogen ion transport in artificial membranes, isolated cell membranes, simple non-epithelial cell systems and various epithelia. Presentations will be organized around the following general topics. I. Theoretical and experimental work on hydrogen ion diffusion in lipid bilayers, cell membranes, surface barriers and paracellular pathways. II. Hydrogen Ion pumps in simple non-epithelial cell systems (bacterial rhodopsin, chloroplasts, mitochondria) and isolated membrane vesicles. III. Electrophysiologic analysis of hydrogen ion transport in various epithelia (stomach, intestine, gall bladder, pancreas, salivary glands, kidney and urinary tract). IV. Structure-function relations, regulatory processes and metabolic coupling of hydrogen ion transport in isolated cells or glands and intact epithelia.

Abstract :
The role of the kidney in the homeostatic control of electrolytes, water and acid-base balance and the derangements that occur in certain pathologic states will be investigated. Metabolic balance, clearance, micropuncture and microperfusion techniques will be utilized in several different species of experimental animals (dogs, rats, rabbits, hamsters, psammomys) to investigate the following problems: H plus secretion---the mechanism of H plus secretion by the tubule, physiologic factors controlling its rate, and nature of the proximal defect in fructuose-induced renal rubular acidosis; proximal NaCl reabsorption and its relation to H plus secretion and reasorption of glucose, phosphate and amino acids; the role of extracellular volume and intrarenal physical factors in regulating distal sodium reasorption; passive model of renal medulla countercurrent system in species other than rabbit.
